<dcterms:abstract xml:lang="en">The development of efficient photosensitizers (PSs) for two-photon photodynamic therapy (2P-PDT) is a step towards a better cancer treatment. The use of fluorescent PSs enables the combination of 2P-PDT&Imaging. In this thesis, we investigated the encapsulation of various unsubstituted fluorenylporphyrins, fluorenylporphyrins bearing electron donor/acceptor groups on their periphery, fluorenylporphyrins capped with ruthenium moieties and fluorenylphthalocyanines into biocompatible polymer-based nanoparticles (NPs) to make these hydrophobic PSs “water-soluble” by putting them into a stable nanosuspension in water. We succeeded in the encapsulation of most of these molecules at higher loading (25wt% relative to polymer mass) compared to the previously reported results (10wt%). Most of the porphyrins remained fluorescent in water while encapsulated and exhibited high two-photon absorption cross-sections (measured via Two Photon Excited Fluorescence), validating their potential for 2P-PDT&Imaging. For the nonfluorescent ruthenium-containing porphyrins, we explored their potential for PDT combined with photothermal therapy. Most of the encapsulated compounds were confirmed to generate singlet oxygen, indicating effective photosensitizing activity. Preliminary in vitro experiments on human breast cancer cells further demonstrated that the photophysical properties correlated well with the observed biological effects, validating our characterization approach for selecting promising candidates for 2P-PDT&Imaging.</dcterms:abstract>
