     <dcterms:abstract xml:lang="fr">Cette thèse explore les mécanismes de résistance aux inhibiteurs de BRAF dans le mélanome muté BRAF, en se focalisant sur le rôle des circARN. Grâce à des analyses transcriptomiques intégratives et à l’outil CirScan, des réseaux circARN-miARN-ARNm impliqués dans la résistance ont été identifiés. Parmi eux, les circARN tels que hsa_circ_0001610 et hsa_circ_0002805 agissent comme des éponges à miARN, séquestrant des miARN clés comme miR-29a-3p et miR-151a-3p. Cette séquestration favorise la surexpression de gènes critiques comme AhR, AXL et EGFR, responsables de la résistance et de l’agressivité tumorale. Les validations fonctionnelles in vitro montrent que la déplétion stable de ces circARN entraîne une restauration de la sensibilité aux inhibiteurs de BRAF et une réduction significative des capacités invasives des cellules tumorales. Ces résultats révèlent le rôle crucial des circARN dans la plasticité cellulaire et dans l’acquisition de la résistance thérapeutique, ouvrant la voie à de nouvelles stratégies thérapeutiques ciblant ces ARN non codants pour améliorer l’efficacité des traitements dans le mélanome métastatique. </dcterms:abstract>
     <dcterms:abstract xml:lang="en">This thesis investigates the mechanisms of resistance to BRAF inhibitors in BRAF-mutated melanoma, focusing on the role of circRNAs. Through integrative transcriptomic analyses and the use of the CirScan tool, circRNA-miRNA-mRNA networks involved in resistance were identified. Among these, circRNAs such as hsa_circ_0001610 and hsa_circ_0002805 act as miRNA sponges, sequestering key miRNAs like miR-29a-3p and miR-151a-3p. This sequestration promotes the overexpression of critical genes such as AhR, AXL, and EGFR, which are responsible for resistance and tumor aggressivenes. Functional validations in vitro demonstrate that stable depletion of these circRNAs restores sensitivity to BRAF inhibitors and significantly reduces the invasive capacities of tumor cells. These findings highlight the crucial role of circRNAs in cellular plasticity and the acquisition of therapeutic resistance, paving the way for new therapeutic strategies targeting these non-coding RNAs to enhance the effectiveness of treatments in metastatic melanoma.</dcterms:abstract>
