     <dcterms:abstract xml:lang="fr">Le 17β-oestradiol (E2) est une hormone pléiotropique issue de l’aromatisation de la testostérone par l’aromatase. Dans le cerveau des mammifères et oiseaux, l’E2 influence comportement et neurogenèse en modulant les neurotransmetteurs. Chez les poissons téléostéens, qui maintiennent une neurogenèse étendue à l’âge adulte, deux gènes codent l’aromatase : cyp19a1b (cérébrale) et cyp19a1a (gonadique). L’aromatase cérébrale est régulée par l’E2 et est fortement exprimée dans les cellules gliales radiaires, qui sont pour certaines progénitrices. Cela suggère un rôle clé de l’aromatase cérébrale dans la neuroplasticité mais le lien causal reste à être investigué. Ce travail a exploré le rôle de l’aromatase cérébrale chez le poisson zèbre, via une mutation ciblant le gène cyp19a1b, et les effets à long terme d’expositions précoces à deux perturbateurs endocriniens : Le 17α-éthinyloestradiol (EE2) et le clotrimazole (CLO). La mutation et les expositions ont modifié divers comportements, comme la nage et la sociabilité, parfois de façon sexe-spécifique à l’âge adulte. Par immunofluorescences, il a été montré que l’aromatase, l’EE2 et le CLO modulent la prolifération cellulaire. Le clotrimazole augmente le nombre de neurones dopaminergiques dans l’hypothalamus caudal, tandis que la mutation accroît la dégradation de la dopamine. Des analyses transcriptomiques chez la lignée mutante ont révélé de nombreux processus biologiques impactés, particulièrement chez les femelles adultes. Ces résultats soulignent le rôle clé de l’aromatisation dans la régulation du comportement et de la neuroplasticité, ainsi que les impacts de sa modulation enzymatique chez le poisson zèbre. </dcterms:abstract>
     <dcterms:abstract xml:lang="en">17β-estradiol (E2) is a pleiotropic hormone derived from the aromatization of testosterone by aromatase. In the brains of mammals and birds, E2 influences behavior and neurogenesis by modulating neurotransmitters. In teleost fish, which maintain extensive neurogenesis into adulthood, two genes encode aromatase : cyp19a1b (brain) et cyp19a1a (gonad). Cerebral aromatase is regulated by E2 and is highly expressed in radial glial cells, some of which are progenitor cells. This suggests a key role for brain aromatase in neuroplasticity, but the causal link remains to be investigated. This work explored the role of brain aromatase in zebrafish, via a mutation targeting the cyp19a1b gene, and the long-term effects of early exposure to two endocrine disruptors : 17α-ethinylestradiol (EE2) and clotrimazole (CLO). The mutation and exposures modified various behaviours, such as swimming and sociability, sometimes in age-specific in adulthood. By immunofluorescences, it was shown that aromatase, EE2 and CLO modulate cell proliferation. Clotrimazole increases the number of dopaminergic neurons in the caudal hypothalamus, while mutation increases dopamine degradation. Transcriptomic analyses in the mutant line revealed numerous biological processes impacted, particularly in adult females. Theses results underline the key role of aromatization in the regulation of behavior and neuroplasticity, and the impact of its enzymatic modulation in zebrafish. </dcterms:abstract>
