     <dcterms:abstract xml:lang="fr">Ce travail de thèse se propose de développer des nanosondes luminescentes pour l’analyse et le tri de sous population de vésicules extracellulaires impliquées dans des pathologies reliées à l’infertilité. Les EV, bicouches lipidiques contenant des informations qui permettent la communication entre les cellules et sont impliquées dans de nombreuses pathologies peuvent rendre compte de l’infertilité d’une patiente. Plus précisément, nous étudierons l’influence de l’insuffisance ovarienne précoce et du syndrome de l’ovaire polykystique sur la morphologie, la taille et le contenu en miRNA des EV. Pour analyser et marquer le contenu, nous proposons de développer des nanosondes luminescentes sensibles à l’environnement intra vésiculaire des EV in situ. Les nanoclusters d’or sont des nanoparticules duales (fluorescentes et à lourd noyau) et biocompatibles de taille inférieure à 2 nm. Leur fluorescence provient aussi de leurs ligands, pour lesquels nous avons élaboré une plateforme chimique, ce qui les rend sensibles à l’environnement. De plus, les nanoclusters offrent des propriétés optiques uniques, une intensité d’émission et un long temps de vie (microseconde) modulables en fonction de l’environnement. Nous avons développé une sonde de pH duale (en intensité d’émission et en temps de vie de fluorescence), qui permet de s’affranchir des problèmes d’autofluorescence des milieux et d’une calibration. </dcterms:abstract>
     <dcterms:abstract xml:lang="en">In this work we intend to develop luminescent nanoprobes to analyze and sort sub populations of extracellular vesicles involved in infertility pathologies. EV have information for in between cell communication encapsulated in lipid bilayers and are implicated in several pathologies such as infertility. We will study how polycystic ovarian syndrome and Ovarian Failure impact EV’ morphology, size and miRNA content. We will develop environment sensible luminescent nanoprobes to analyze and label the content of EV in situ. Gold nanocluster are biocompatible duals probes (fluorescent and with heavy core) with a size under 2 nm. Their luminescent properties are conferred partly by their ligand which can be chemically tuned and therefore are sensible to their environment. Moreover, they have unique optical properties: emission intensity and long microsecond lifetimes that are environment tunable. We have developed a dual (fluorescence lifetime and intensity) pH probe that help prevent sample’s auto-fluorescence nanoprobe interference and need no calibration.</dcterms:abstract>
