     <dcterms:abstract xml:lang="fr">Le carcinome hépatocellulaire (CHC), le principal cancer primitif du foie, reste difficile à détecter aux stades précoces d’où la nécessité de concevoir de nouveaux outils de détection, par exemple par une approche de vectorisation. Nous avons sélectionné deux peptides présentant un tropisme hépatocytaire : le GBVA10-9, inhibant partiellement l’infection par le virus d’hépatite C, et le peptide CPB qui correspond à un fragment de la protéine circumsporozoïte de Plasmodium berghei impliquée dans l’infection hépatocytaire. Nous les avons radiomarqués au cuivre-64 et au gallium-68 et étudié leur tropisme vers des cellules d’hépatomes humains et murin in vitro et dans un modèle de CHC chez la souris.Nous avons montré que le GBV10-9 radiomarqué est mieux capté par les cellules d’hépatome par rapport au CPB. Parallèlement, j’ai caractérisé la captation de nanoparticules (NPs) polymères formulées à partir de l’homopolymère poly(malate de benzyle) (PMLABe73) et/ou de son dérivé amphiphile pegylé (PEG42-b-PMLABe73) par les cellules hépatiques, les macrophages et les cellules mononucléées du sang périphérique puis étudié l’impact de la fonctionnalisation des NPs par le peptide GBVA10-9 sur cette captation.</dcterms:abstract>
     <dcterms:abstract xml:lang="en">Hepatocellular carcinoma (HCC), the main primary liver cancer, remains difficult to detect at early stages, hence the need to design new detection tools, for example using a vectorization approach. We selected two peptides presenting a hepatotropism: GBVA10-9 peptide, able to partially inhibit infection by the hepatitis C virus, and the CPB peptide which corresponds to a fragment of the circumsporozoite protein of Plasmodium berghei involved in hepatocyte infection. We radiolabeled them with copper-64 and gallium-68 and studied their tropism towards human and murine hepatoma cells in vitro and in a mouse model of HCC. We showed that radiolabeled GBV10-9 is better internalized by hepatoma cells compared to CPB. At the same time, I characterized the uptake of polymer nanoparticles (NPs) formulated from the poly(benzyl malate) homopolymer (PMLABe73) and/or its pegylated amphiphilic derivative (PEG42-b-PMLABe73) by liver cells, macrophages and peripheral blood mononuclear cells then studied the impact of the functionalization of NPs by GBVA10-9 peptide on this uptake.</dcterms:abstract>
