     <dcterms:abstract xml:lang="fr">Les troubles de la fertilité et les anomalies du tractus uro-génital sont en augmentation. Afin de mieux comprendre ce déclin de la santé reproductive, il est important d’approfondir nos connaissances sur la gamétogenèse et sur le développement et la physiologie des gonades car nombre de ces anomalies tirent leur origine au cours du développement. Dans un premier axe de recherche, j’ai activement contribué la compréhension de la transition mitose-méiose, au cours de la spermatogénèse. Mes travaux ont permis la caractérisation à l’échelle de la cellule unique d’un troisième mécanisme d’inactivation transcriptionnelle pangénomique retrouvé en début de prophase I de méiose chez la souris appelé Early Meotic Low Transcription (EMLT). J’ai ainsi démontré que ce mécanisme est conservé au cours de l’évolution chez les mammifères et qu’il s’accompagne d’une stabilisation des ARNm. Dans un deuxième axe de recherche, j’ai établi une méta-cartographie à l’échelle de la cellule unique des gonades au cours du développement. Ce méta-atlas inter-espèce, composé de plus d’un million de cellules humaines et souris provenant de 25 études, constitue la ressource la plus exhaustive concernant le développement et la physiologie des gonades. Ces données ont été rendues disponibles sur la troisième génération du ReproGenomics Viewer (RGV3, https://rgv.genouest.org). J’ai ainsi contribué à la refonte complète de RGV3 dont la nouvelle interface interactive facilite l’exploration des données de scRNA-seq et est compatible avec les données de transcriptomique spatiale. </dcterms:abstract>
     <dcterms:abstract xml:lang="en">Fertility disorders and genitourinary abnormalities are on the rise. To better understand this decline in reproductive health, it is important to increase our knowledge of gametogenesis and gonadal development and physiology, as many of these abnormalities originate during development. In a first line of research, I have actively contributed to our understanding of the mitosis-meiosis transition during spermatogenesis. My work has led to the single-cell characterisation of a third genome-wide transcriptional inactivation mechanism found at the onset of prophase I of meiosis in the mouse, called Early Meotic Low Transcription (EMLT). I have shown that this mechanism is conserved during mammalian evolution and that it is associated with mRNA stabilisation. In a second line of research, I have assembled a mega single-cell meta-atlas of gonads during development. This cross-species meta-atlas, comprising more than a million human and mouse cells from 25 studies, is the most comprehensive resource on gonadal development and physiology. These data have been made available on the third generation of the ReproGenomics Viewer (RGV3, https://rgv.genouest.org). I have contributed to the complete redesign of RGV3, whose new interactive interface facilitates exploration of scRNA-seq data and is compatible with spatial transcriptomics data.</dcterms:abstract>
