     <dcterms:abstract xml:lang="fr">Près de la moitié des mélanomes cutanés diagnostiqués comportent une altération activatrice sur le codon V600 du gène codant pour la protéine kinase BRAF. Cela a permis l'avènement des inhibiteurs ciblant spécifiquement la forme mutée de la protéine, révolutionnant ainsi le traitement de cette maladie. L'efficacité de ces thérapies ciblées demeure néanmoins limitée par l'émergence rapide de mécanismes de résistance. Cette réalité clinique souligne le besoin urgent de développer de nouvelles stratégies thérapeutiques pour surmonter la résistance et améliorer la survie des patients. Dans cette étude, nous montrons que l'activité mitochondriale, l'accumulation d'espèces réactives de l'oxygène (ROS) et l'augmentation de la peroxydation des lipides conduisent à une plus grande vulnérabilité vis-à-vis de la mort par ferroptose des cellules de mélanome résistantes aux inhibiteurs de BRAF. Ainsi, nous avons pu montrer que la ferroptose constituait une opportunité thérapeutique pour cibler les cellules résistantes, qui présentent une sensibilité accrue aux inducteurs chimiques de cette forme de mort cellulaire. En outre, nos résultats suggèrent fortement que le facteur de transcription Aryl Hydrocarbon Receptor (AhR), dont nous avons déjà montré l’implication dans l’acquisition de la résistance aux thérapies ciblées, joue également un rôle dans l’émergence de cette vulnérabilité à la ferroptose.</dcterms:abstract>
     <dcterms:abstract xml:lang="en">Nearly half of all diagnosed cutaneous melanomas carry an activating alteration at codon V600 of the gene encoding the BRAF protein kinase. This has led to the era of inhibitors specifically targeting the mutated form of the protein, thereby revolutionizing the treatment of this disease. However, the efficacy of these targeted therapies remains limited by the rapid emergence of resistance mechanisms. This clinical reality underlines the urgent need to develop new therapeutic strategies to overcome resistance and improve patient survival. In this study, we show that mitochondrial activity, accumulation of reactive oxygen species (ROS) and increased lipid peroxidation lead to greater vulnerability to the death by ferroptosis in BRAF inhibitor-resistant melanoma cells. Thus, we have shown that ferroptosis represents a therapeutic opportunity to target resistant cells, which show increased sensitivity to chemical inducers of this form of cell death. Furthermore, our results strongly suggest that the transcription factor Aryl Hydrocarbon Receptor (AhR), which we have already shown to be involved in the acquisition of resistance to targeted therapies, also plays a role in the emergence of this vulnerability.</dcterms:abstract>
