     <dcterms:abstract xml:lang="fr">Staphylococcus aureus est un pathogène opportuniste, multi-résistant aux antibiotiques et pouvant causer de sévères infections chez l’Homme. Dans ces travaux de thèse, nous nous sommes intéressés aux toxines SprA1, SprA2, SprG1₃₁ et SprG1₄₄ exprimées par S. aureus. Ces toxines sont de petits peptides cationiques et membranaires capables d’inhiber la croissance de S. aureus en condition de surexpression. Notre but était de mieux comprendre le mécanisme d’action de ces toxines. L’étude des effets de la surexpression de ces peptides sur la membrane a montré que ces trois peptides induisent une dépolarisation de la membrane. Celle-ci est concomitante à la formation de larges pores membranaires pour SprA1, alors que SprA2 forme des pores transitoires et/ou plus petits. SprG1₃₁ et SprG1₄₄ semblent ne pas former de pores, mais induire la formation de mésosomes dans la membrane. Cette étude a été complétée par des analyses des interactions de ces peptides avec des modèles membranaires. Il a ainsi été montré que SprA2 présente une forte sélectivité envers les membranes zwitterioniques et que SprG1₄₄ semble capable d’agréger les érythrocytes. De plus, la présence de SprG1₃₁ et SprG1₄₄ dans les vésicules extracellulaires a été mise en évidence. Ces travaux permettent de mieux comprendre l’action de ces toxines et d’apporter des indices sur leurs fonctions biologiques.</dcterms:abstract>
     <dcterms:abstract xml:lang="en">Staphylococcus aureus is an opportunistic pathogen, multi-resistant to antibiotics, and causing severe infections in humans. During this thesis, we focused on toxins SprA1, SprA2, SprG1₃₁ and SprG1₄₄ expressed by S. aureus. These toxins are small cationic membrane peptides, able to inhibit bacterial cell growth when overexpressed. Our goal was to investigate the mechanism of action of these toxins. The study of the effects of peptides overexpression on S. aureus membrane showed that these three peptides can induce a membrane depolarization. On the same time, SprA1 is able to form large stable pores in the membrane, whereas SprA2 forms smaller and/or transient pores. SprG1₃₁ and SprG1₄₄ do not seem to form pores, but they are able to induce mesosomes-like structures in the membrane. This study has been completed by the investigation of peptide-membrane interactions with model membranes. Thus, our results demonstrate that SprA2 has a high membrane selectivity for zwitterionic membranes and SprG1₄₄ seems to aggregate erythrocytes. Moreover, we showed that SprG1₃₁ and SprG1₄₄ are present in extracellular vesicles. Together, our experiments are providing a better understanding of the mode of action of these toxins and clues to their biological function.</dcterms:abstract>
