     <dcterms:abstract xml:lang="fr">La différenciation des lymphocytes B (LyB) en plasmocytes (PC) est un processus complexe pouvant faire place à l’émergence de maladies malignes lymphoprolifératives. Les enzymes PIM sont des Ser/Thr kinases qui phosphorylent un grand nombre de protéines favorisant ainsi la survie, la prolifération, la migration cellulaire et, dans les cancers, l’agressivité et la résistance au traitement. L’engagement précoce des LyB vers la génération des PC est associé à une importante reprogrammation cellulaire où émerge une forte expression de PIM2. Par son activité kinase, PIM2 exerce un rôle pivot dans le processus de différenciation en favorisant l’entrée en cycle cellulaire et en contrôlant l’activation de la caspase 3. Dans les PC matures, PIM2 se maintient à haut niveau et perpétue sa fonction dans le contrôle de l’apoptose. Par son action directe sur la réponse au stress cellulaire du RE, elle atténue l’activation de la voie de l’ISR dépendante de la kinase PERK, ce qui contribue au contrôle de la voie mitochondriale de l’apoptose. Par nos approches nous montrons une convergence des effets médiés par PIM2 sur cette voie en particulier en diminuant la charge de MCL1 en molécules pro-apoptotiques. Dans le myélome, les PC tumoraux sont dépendants de PIM2 au même titre que MCL1 pour leur survie. Sur la base de ces résultats, nous montrons que l’inhibition combinée de PIM2 et MCL1 dans des lignées de myélome est fortement synergique pour induire la mort des cellules in vitro. In vivo, dans notre modèle de xénogreffe original, cette association thérapeutique permet un blocage de la progression tumorale.</dcterms:abstract>
     <dcterms:abstract xml:lang="en">The differentiation of B cells into plasma cells (PC) is a complex process that can give rise to the emergence of malignant lymphoproliferative diseases. PIM enzymes are Ser/Thr kinases that phosphorylate a large number of proteins, thereby promoting survival, proliferation, cell migration and, in cancers, aggressiveness and resistance to treatment. The early commitment of B cells towards PC generation is associated with extensive cellular reprogramming where emerge a strong increase in the expression of PIM2. Through its kinase activity, PIM2 play a pivotal role in the differentiation process, facilitating cell cycle entry and the control of caspase 3 activation. In mature PC, PIM2 is maintained at high level and perpetuates its role in the control of apoptosis. Through its direct action on the ER cellular stress response, it attenuates activation of the PERK kinase-dependent ISR pathway, which contributes to control the mitochondrial pathway of apoptosis. Our approaches show a convergence of PIM2-mediated effects on this pathway, in particular by reducing the load of pro-apoptotic molecules on MCL1. In myeloma, tumor PC are dependent on PIM2 as well as MCL1 for survival. Based on these results, we show that combined inhibition of PIM2 and MCL1 in myeloma cell lines is highly synergistic in inducing cell death in vitro. In vivo, in our original xenograft model, this therapeutic combination blocks tumor progression.</dcterms:abstract>
