     <dcterms:abstract xml:lang="fr">Ces dernières années, de considérables progrès ont été obtenus en médecine génomique. L’apport de la technologie de séquençage haut-débit dans les laboratoires de génétique médicale a notamment permis de mieux diagnostiquer les pathologies génétiques rares. Malgré cette avancée, un nombre important de patients diagnostiqués se retrouvent en errance thérapeutique. Environ 250 millions de personnes souffrent de pathologies génétiques rares et moins de 5% de ces pathologies disposent aujourd’hui de traitements efficaces. La découverte et l’amélioration constante des outils CRISPR/Cas9 ont ouvert la porte à une nouvelle médecine, la chirurgie du génome. Dans ce contexte, une partie importante des altérations génétiques responsables de maladies rares peuvent être en théorie directement corrigées par cette approche, ouvrant une perspective thérapeutique inespérée pour des patients étant parfois les seuls porteurs de ces altérations génétiques. Au cours de ma thèse, j’ai l’opportunité de développer et d’évaluer les outils d’édition du génome CRISPR/Cas9, d’une part pour concevoir et mesurer l’efficacité de nouveaux traitement in vitro, et d’autre part pour installer et décrypter le rôle biologique de variants génétiques de signification inconnue. Désormais, il est possible de corriger ou créer ces variants en recherche fondamentale afin d’aider les cliniciens à conclure sur leur rôle biologique ouvrant la porte à des thérapies personnalisées dans un futur proche.</dcterms:abstract>
     <dcterms:abstract xml:lang="en">Recently, considerable progress has been made in genomic medicine. The use of high-throughput sequencing technology in medical genetics laboratories has made it possible to diagnose rare genetic pathologies. Conversely, a significant number of diagnosed patients are in treatment wandering. No less than 250 million people suffer from rare genetic diseases and less than 5% of these diseases have effective treatments today. Thanks to the emergence and constant improvement of CRISPR/Cas9 tools, medicine has entered a new therapeutic era of genome surgery. In this context, an important part of the genetic alterations responsible for rare diseases can, in theory, be directly corrected by this approach, opening an unexpected therapeutic perspective for patients being sometimes the only carriers of these genetic alterations. During my thesis, I had the opportunity to develop CRISPR/Cas9 gene editing tools, first to design and measure the efficiency of new treatments in vitro, and second to install and decipher the biological role of variants of unknown significance. It is now possible to correct or create these variants in basic research in order to help clinicians conclude their biological role, opening the door to personalized therapies in the near future.</dcterms:abstract>
