     <dcterms:abstract xml:lang="fr">L’épithélium intestinal et le microbiote intestinal sont en équilibre dans l’intestin. Cependant, cet équilibre peut être rompu par différents facteurs endogènes ou exogènes, conduisant à la dysbiose intestinale. La dysbiose intestinale est une perturbation de la composition du microbiote intestinal, pouvant être corrélée à l’apparition de certaines pathologies intestinales, telles que la maladie de Crohn, l’obésité, le diabète de type 2 ou le cancer colorectal. Certaines bactéries comme Salmonella sont augmentées et persistent dans l’intestin tandis que d’autres espèces bactériennes commensales sont diminuées, telles que Bacteroides fragilis et Roseburia intestinalis, considérées comme des potentiels probiotiques de nouvelle génération. Ces potentiels probiotiques de nouvelle génération sont capables de produire des métabolites bactériens exerçant des effets bénéfiques dans l’intestin et d’autres organes. Dans un premier temps, nous avons montré que les métabolites, comprenant l’acide cholique, sécrétés par B. fragilis étaient capables d’inhiber la virulence de Salmonella Heidelberg dans des modèles in vitro et in vivo. Dans un deuxième temps, nous avons observé que R. intestinalis produisait des métabolites capables de stimuler l’expression de PYY, peptide hormonal, sur un modèle multi- cellulaire comprenant des cellules entéro- endocrines. Ces deux études montrent l’importance des métabolites produits par le microbiote intestinal, pour le maintien de l’intégrité intestinal. L’administration des probiotiques de nouvelle génération et/ou de leurs métabolites pourraient conduire à la mise en place de nouveaux traitements, dans les cas de pathologies associées aux dysbioses. Cependant, de nombreux mécanismes sont encore à élucider, notamment l’étude des communautés bactériennes et les études cliniques qui sont en cours d’élaboration.</dcterms:abstract>
     <dcterms:abstract xml:lang="en"> The intestinal epithelium and the intestinal microbiota are in balance in the intestine. However, this balance can be disrupted by various endogenous or exogenous factors, leading to intestinal dysbiosis. Intestinal dysbiosis is a disturbance of the composition of the intestinal microbiota, which can be correlated to the onset of certain intestinal pathologies, such as Crohn's disease, obesity, type 2 diabetes or colorectal cancer. Some bacteria such as Salmonella are increased and persist in the intestine while other commensal bacterial species are decreased, such as Bacteroides fragilis and Roseburia intestinalis, which are considered to be potential next generation probiotics. These potential next generation probiotics are able to produce bacterial metabolites with beneficial effects in the gut and other organs. First, we showed that the metabolites, including cholic acid, secreted by B. fragilis were able to inhibit the virulence of Salmonella Heidelberg on in vitro and in vivo models. In a second step, we observed that R. intestinalis produced metabolites is able to stimulated the expression of PYY, a hormonal peptide, in a multi-cellular model including enteroendocrine cells. These two studies show the importance of the metabolites produced by microbiota, for the maintenance of the intestinal integrity. The administration of next generation probiotics and/or their metabolites could lead to the implementation of new treatments for pathologies associated with dysbiosis. However, many mechanisms are still to be elucidated, including the study of bacterial communities and clinical studies.</dcterms:abstract>
