     <dcterms:abstract xml:lang="fr">Le mélanome est la forme la plus meurtrière des cancers cutanés. Dans près de la moitié des cas, les cellules tumorales portent la mutation BRAFV600, à l’origine d’une activation constitutive de la voie des MAPK favorisant la croissance tumorale. La présence de cette mutation rend les patients éligibles aux thérapies ciblées : les inhibiteurs de BRAF (BRAFi). L’utilisation des BRAFi, en combinaison avec les inhibiteurs de MEK engendre rapidement une réponse impressionnante chez les patients. Malheureusement, dans la majorité des cas, des mécanismes de résistance se développent et aboutissent à la rechute des patients. L’hétérogénéité tumorale et la grande plasticité dont les cellules de mélanome font preuve leur permettent de s’adapter rapidement au traitement suite à l’émergence progressive de cellules tumorales résistantes. Des travaux préalables réalisés au laboratoire avaient démontré que l’activation canonique d’AhR induit l’expression de gènes impliqués dans l’acquisition de la résistance aux BRAFi. Nous rapportons maintenant que l’activation d’AhR entraine l’activation de son partenaire, l’oncoprotéine Src. L’activation de cet axe AhR/Src induit une reprogrammation des cellules de mélanome vers un état phénotypique agressif, invasif et dédifférencié. A l’inverse, l’inhibition de l’axe AhR/Src permet de reverser les cellules vers un programme de sensibilité. Nous avons également montré, via un modèle PDX de tumeurs résistantes de mélanome, que l'utilisation d’inhibiteurs de Src permet de resensibiliser les cellules de mélanome à l'inhibition de BRAF. L'ensemble de ces travaux met en évidence l’espoir d’une nouvelle option thérapeutique pour les patients.</dcterms:abstract>
     <dcterms:abstract xml:lang="en">Melanoma is the deadliest form of skin cancer. In half of all cases, tumor cells carry the BRAFV600E mutation, which causes an uncontrolled activation of the MAPK pathway and promotes tumor growth. The presence of this mutation makes patients eligible for targeted therapies: the BRAF inhibitors (BRAFi). The use of BRAFi induces an impressive response in patients. Unfortunately, in the most cases, melanoma cells become resistant leading to patient relapse. The tumor heterogeneity and the high plasticity of melanoma cells contribute to the emergence of resistant cells. We implicate the transcription factor AhR in the acquisition of resistance. Indeed, canonical activation of AhR induces the expression of genes associated with the acquisition of resistance to BRAFi. Moreover, AhR activation leads to the activation of its protein partner Src resulting in the acquisition of an aggressive invasive and dedifferentiated phenotype of melanoma cells. Conversely, inhibition of the AhR/Src axis allows the cells to be reversed to a sensitive program. The therapeutic use of Src inhibitors allows resensitization of BRAFi-resistant melanoma cells, and could potentially delay the development of resistance and the occurrence of relapse of the patients.</dcterms:abstract>
