     <dcterms:abstract xml:lang="fr">Le virus Chikungunya (CHIKV) est un virus myotrope qui a provoqué depuis sa réapparition en 2004 dans l'océan Indien, des millions de cas d'arthrite et de maladies musculo-squelettiques en Europe, en Asie et en Amérique. Nous avons identifié (par criblage CRISPR-Cas9) la protéine Four- and-a-Half LIM protein 1 (FHL1) comme un facteur hôte majeur pour la réplication et la pathogenèse de CHIKV. FHL1 est fortement exprimé dans les muscles striés et les fibroblastes, deux cibles principales du CHIKV. Bien que sa fonction physiologique soit mal connue, FHL1 est essentiel à la myogenèse. Nous montrons que les fibroblastes et les cellules musculaires issus de patients atteints de dystrophie musculaire Emery-Dreifuss hébergeant des mutations dans le gène Fhl1 entraînant une absence de protéine FHL1, sont résistants à l'infection. Des expériences in vivo démontrent que les souris dépourvues de FHL1 sont résistantes à l'infection et n'ont montré aucune pathologie détectable, indiquant que FHL1 est un facteur majeur de permissivité cellulaire à l'infection à CHIKV.</dcterms:abstract>
     <dcterms:abstract xml:lang="en">Chikungunya virus (CHIKV) is a myotropic virus, which has caused since its re-emergence in 2004 in the Indian Ocean, millions of cases of debilitating arthritis and musculoskeletal disease into Europe, Asia, and the Americas. We identified (by CRISPR-Cas9 screen) the Four-and-a-Half LIM protein 1 (FHL1) as a major host factor for CHIKV replication and pathogenesis. FHL1 is strongly expressed in striated muscle and fibroblasts, two major CHIKV targets. Although its physiological function is poorly understood, FHL1 is essential for muscle development (myogenesis). We show that fibroblasts and muscle cells derived from Emery-Dreifuss muscular dystrophy patients harboring mutations in the FHL1 gene resulting in the absence of FHL1 protein, are resistant to CHIKV infection. In vivo experiments demonstrate that mice lacking FHL1 are resistant to CHIKV infection and showed no detectable pathology, indicating that FHL1 is a major factor for cell permissiveness to CHIKV infection.</dcterms:abstract>
