     <dcterms:abstract xml:lang="fr">Le poly(acide malique) et ses dérivés sont des polymères aux propriétés de biocompatibilité et de bio(dégradabilité) remarquables, qui en font des candidats de choix pour l'élaboration de systèmes de vectorisation de principes actifs. Une première partie de ce doctorat a été consacrée à l’évaluation de la biocompatibilité et de la captation de nanoparticules (NPs) fluorescentes formées à partir de poly(malate de benzyle) (PMLABe) obtenus par polymérisation soit chimique soit enzymatique du malolactonate de benzyle par des cellules hépatiques HepaRG et des macrophages humains primaires. .	Toutes les NPs sont peu toxiques, mais les niveaux de captation par les cellules HepaRG et les macrophages varient considérablement en fonction des caractéristiques physico-chimiques des NPs. Un second objectif était d’évaluer l’influence du taux de PEGylation de NPs à base de PMLABe et de PEG-b-PMLABe sur leur captation cellulaire. Enfin, nous avons évalué la biocompatibilité et la captation de NPs à base de dérivés hydrophobes et amphiphiles du poly(acide diméthylmalique) : les niveaux de toxicité et d’internalisation semblent être relatifs aux caractéristiques physico-chimiques des NPs.</dcterms:abstract>
     <dcterms:abstract xml:lang="en">Poly(malic acid) and its derivatives are polymers with outstanding biocompatibility and bio(degradability) properties, which make them promising candidates for the development of drug delivery systems. The first part of this thesis was devoted to the evaluation of the biocompatibility and the uptake of fluorescent nanoparticles (NPs) formed from poly(benzyl malate) (PMLABe), obtained by either chemical or enzymatic polymerization of benzyl malolactonate, by HepaRG hepatoma cells and primary human macrophages.	All NPs are not very toxic, but the levels of uptake by HepaRG cells and macrophages vary considerably depending on the physicochemical characteristics of NPs. A second objective was to evaluate the influence of the PEGylation rate of NPs based on PMLABe and PEG-b-PMLABe on their cell uptake. Finally, we evaluated the biocompatibility and the internalization of NPs based on hydrophobic and amphiphilic derivatives of poly(dimethylmalic acid): the levels of toxicity and internalization seem to be depending on the physicochemical characteristics of NPs.</dcterms:abstract>
