Elle affecte le développement précoce et est principalement due à un déficit en Sonic Hedgehog (SHH). Les patients portant des formes peu sévères sont viables mais présentent souvent des malformations craniofaciales.. Un modèle souris, Shh+/- , a été généré et son phénotype étudié. Une malformation discrète de la glande hypophysaire et une anomalie du basisphénoide ont été observées chez ces mutants. Cette malformation est due à une persistance du canal buccohypophyseal, au niveau de la ligne médiane. Ce mutant constitue une excellente opportunité en orthodontie pour étudier la croissance craniofaciale en présence d’une anomalie congénitale du basisphénoide. Cette étude a montré qu’une fusion prématurée de la synchondrose sphéno-occiptale était observée chez les mutants Shh+/- adultes ; ce qui provoque des problèmes de croissance du crâne et sa courbure. Ce travail montre également que les patients présentant un diagnostic moléculaire suggérant une déficience en signal SHH doivent être suivis en orthodontie. Holoprosencephaly (HPE) is a pathology which affects the formation of the brain and the face. It takes place during early development and is mainly due to a deficit in Sonic Hedgehog (SHH) signaling. Patients with mild forms are viable but frequently have craniofacial malformations. A mouse model, Shh+/- was generated and its phenotype studied. I have observed a discrete malformation of the pituitary gland associated with a basisphenoid anomaly. This malformation is due to the persistence of the buccohypophyseal canal at the level of the midline. This mutant is an excellent opportunity for orthodontics to study craniofacial growth in the presence of a congenital abnormality of the basisphenoid. This study have revealed that a premature fusion of spheno-occipital synchondrosis was observed in Shh+/- adult mutants, leading to skull growth problems and its curvature. This work also shows that patients with a molecular diagnosis exhibiting a SHH-deficiency should be followed by orthodontics.
